You can manualyy update the locate database at any time by running the
command "updatedb" (as root).
A standard RH install sets up a cron job to update the locate database
every day (at some time early in the morning). If your computer is
never on early in the morning, you can edit the crontab to that the
database gets updated at a time when your computer is usually going to
be on (see the manpage for crontab).
Another option is to use anacron instead of cron. Anacron was designed
for machines, like laptops, which are not up 24/7.
